Asiaticoside Introduction
What is Asiaticoside?
Asiaticoside is a primary saponin group of phytochemicals present in Centella Asiatica. Centella Asiatica is commonly known as Gotu Kola. This is a popular medicinal plant that serves multiple health benefits. A standardized extraction process is followed to extract Asiaticoside. Asiaticoside has different synonyms like Asiaticosid, Emdecassol, Dermatologico. [1] The molecular formula of Asiaticoside is C48H78O19, CAS registration number is 16830-15-2 and the molecular weight is 959.1. Asiaticoside is a crystalline solid that dissolves in organic solvents like methanol, ethanol, and pyridine that are liquidated in presence of inert gas. [2]
Asiaticoside sources
- Centella Asiatica is an herbal plant with several local names. In Ayurveda, Centella Asiatica is well known as mandukparni or Indian pennywort or jalbrahm. In China, Centella asiatica is famous as gotu kola. Chinese people believe Centella asiatica is a miracle elixir of life. [3] Centella Asiatica belongs to the Umbelliferae family. Hydrocotyle Asiatica is synonymously used for Centella asiatica. [4] Leaves of the Centella Asiatica plant are small, green, and fan-shaped. Centella Asiatica has white or light purple-to-pink flowers and tiny oval-shaped fruits.
- The whole Centella asiaticaplant has plenty of medicinal values and acts as a blood purifier, anti-hypertensive, memory booster, nerve and brain tissue vitalizing agent, anti-depressant, and promoting longevity. However, Centella Asiatica started getting attention in Western medicine in the middle of the twentieth century after a medical report documented that alcohol extracts of Centella Asiatica have positive results against leprosy. [3]
- Areas surrounded by water bodies are favorable for the growth of Centella Asiatica. Widely growing swampy lands of India, Pakistan, Madagascar, Eastern Europe, Sri Lanka, the Southern part of Africa, and the South Pacific are ideal for Centella Asiatica [3] Centella Asiatica also abundantly grow at 650 meters altitude of China and Vietnam. [5]
Constituents of Centella Asiatica extract
Centella Asiatica is composed of centelloids, which are chemically known as ursane-type pentacyclic triterpenoids. The main active ingredients included in the centelloids are asiaticoside, madecasosside (brahminoside), asiatic acid, and madecassic acid (brahmic acid). Other triterpenoids included active constituents in Centella asiatica are: asiaticoside C, D, E, F; centellasaponin B, C; isothankunic acid and oleanane type saponins, e.g. terminolic acid; centellasaponinD. C. asiatica contains about 0.1% essential oils with α-humulene, germacrene B/D, β-caryphyllene, flavonoids, sesquiterpenes, and steroids. The range of Saponins present in Centella is in between 1% to 8%. European Pharmacopoeia specifically mentioned that Centella asiatica contains not less than 6.0% of Saponin contents. [4]
Researchers had estimated three different methanolic extract samples of Centella Asiatica contain 2.82%, 2.68%, and 2.8% of Asiaticoside. Also, researcher’s assessed that three different ethanolic extract samples of Centella Asiatica contained 2.79%, 2.75%, and 2.91% respectively. [6]

How does Asiaticoside work
- Mechanism of action
Asiaticoside inhibits inflammatory responses by inhibiting the inflammatory signaling by downregulating multiple inflammatory mediators like TNF-α, IL-6, etc. [7]
Asiaticoside increases hydroxyproline content and tensile strength. Asiaticoside also enhances the type I and type III procollagen mRNA level and proteins synthesis that supports collagen synthesis. asiaticoside causes stimulation of vascular endothelial growth factor production that supports keratinocytes enhancement. Asiaticoside present has anti-proliferative action at a high concentration that prevents cancerous skin cell growth. [4]
Asiaticoside, asiaticoside a and b, madecassoside and brahminoside stimulate lipolysis and blood microcirculation. These phytochemicals of Centella Asiatica have an inhibitory effect on local adiposity or cellulite. [8] Thus, it helps in weight management.
Asiaticoside manufacturing process
The asiaticoside is obtained from Centella Asiatica plant by using different extraction methods. Following are some standardized extraction processes used to extract asiaticoside. [5]
Liquid solvent extractionÂ
Centella Asiatica has dissolved ethanol/water/ethanol solution in a ratio of 3: 100. For instance, take 3 gm. of Centella Asiatica and ethanol/water/ethanol in a vessel that has attached propeller to make a homogeneous solution. After adding these raw ingredients, the vessel continuously is stirring with 1200 RPM for 24 hrs for extracting asiaticoside from Centella Asiatica and dissolves into that solvent. [5]
Â
Heat flux extraction
In this process, prepare the solvent by mixing methanol (64°C), water (100°C), and Ethanol (78°C) in a Vessel. Then add the required amount of Centella Asiatica in the vessel. The solution is then the heat for 5 hours for ouasiaticoside extraction. [5]
Soxlet extraction
Prepare a solvent by mixing methanol, ethanol, and water (100ml) and add 3gm of Centella Asiatica into it. The solution is then boiled for 5 hours at the boiling point of each solvent. [5]
Ethanolic extract of asiaticoside
The aerial part of Centella Asiatica is dried at 40°C temperature. Then grind them to make a coarse powder. The herb powder is then macerated with 95% ethanol for three days. Then the supernatant liquid is collected. Put into a proper vessel for evaporation of the solvent. After evaporation, the residual asiaticoside extract is stored at -20°C. [5]
Asiaticoside Uses
Asiaticoside in skincare (healing for wounds and burns)
Asiaticoside and other triterpenoids present in C. Asiatica have a wound-healing function. Topically applied Asiaticoside containing formulation can heal the wound without causing skin irritation. Application of Asiaticoside formulation forms a thick epithelial layer and keratin along with the moderate amount of collagen, fibroblasts, and granulation tissue formation over the skin wound area. [9]
Researchers had specifically mentioned that asiaticoside and madecassoside help in burn wound healing by improving collagen I production. Therefore, asiaticoside obtain from C. Asiatica extracts is an effective treatment for both incision and burn wounds. [10]
Asiaticoside breastfeeding
Asiaticoside is one of the primary bioactive compounds of Centella Asiatica. Centella Asiatica is effective to treat stretch marks present in the breast. Application of Centella Asiatica formulation at an early stage of pregnancy can prevent stretch marks. A study conducting on 25 females reported that Centella Asiatica scored the highest in preventive action against stretch marks. [11] It has also been found that Centella Asiatica improves breast tissue elasticity. [12]
Asiaticoside and wrinkles
Wrinkles occur because of lack of elasticity and dehydration in the cellular structure of the skin. Asiaticoside containing Centella Asiatica shows synergistic antioxidant, anti-inflammatory, and UV protective properties with other multiple herbal ingredients, including aloe vera, turmeric, cinnamon barks, etc. The research result showed that a combination of an herbal preparation containing Centella Asiatica improves skin extensibility, firmness, and improved skin hydration. [13]
Asiaticoside vein study
The positive angiogenesis induced by asiaticoside causes stimulation of vascular endothelial growth factor production and resultant of this leads to monocyte chemotactic protein-1 (MCP-1) expression enhancement in keratinocytes and interleukin-1β (IL-β) in macrophages. [13]
Buy lipstick with asiaticoside
Asiaticoside lipsticks give extra care by preventing lip wrinkles. [14]
Antidepressant like effect of asiaticoside

Asiaticoside follows different mechanisms of action to provide multiple health benefits. It has been found that asiaticoside helps in neuron survival, neurotransmitter regulation, and neuron maintenance by increasing brain-derived neurotrophic factor (BDNF). This mechanism of action Asiaticoside exerts therapeutic benefits in patients with neurodegenerative diseases. [7]
Cardiovascular benefit
Asiaticoside has cardiovascular benefits. In an animal, research results found that asiaticoside reduced mean pulmonary hypertension at the and right ventricular hypertrophy in hypoxic conditions. Asiaticoside increases Nitric oxide production that positively impacts endothelial cells and protects pulmonary hypertension. Asiaticoside is effective to inhibit inflammatory signaling pathways by inhibiting the overexpression of different inflammatory mediators. Asiaticoside gives protection from endothelial permeability by reducing endothelial permeability and vascular cell adhesion. [7]
Respiratory diseases
Asiaticoside has anti-inflammatory property that inhibits the inflammatory response in lung tissue. [7]
Bone disorder
Asiaticoside has therapeutic benefits against bone disorder by inducing osteoclasts, and regeneration of periodontal tissue. [7]
Asiaticoside safety
FDA analyzed ceratin case studies and found that the topical application of Madecassol which is an extract of Gotu kola contains madecassic acid, Asiatic acid, and asiaticoside could cause irritant reactions. [15]
Oral supplementation of asiaticoside extracted from C. Asiatica with a dose of 1g per kg body weight has no toxic benefit and is well tolerated by patients. [8]
Toxicity study and cutaneous hypersensitivity test results show that the application of Centella Asiatica is harmless in both systemic and topical use. [16]
Asiaticoside side effects
A high dose of transdermal administration of asiaticoside does not cause any systematic toxicity. But excessive keratinization on the skin at the application site is noticeable. [7]
