 Madecassoside Introduction

What is Madecassoside Powder?

Madecassoside is a bioactive triterpene that is primarily obtained from Centella Asiatica plant extract. Asiaticoside A is another name for madecassoside. Chemically, madecassoside is also known as [6-[[3,4-dihydroxy-6-(hydroxymethyl)-5-(3,4,5-trihydroxy-6-methyloxan-2-yl)oxyoxan-2-yl]oxymethyl]-3,4,5-trihydroxyoxan-2-yl] 8,10,11-trihydroxy-9-(hydroxymethyl)-1,2,6a,6b,9,12a-hexamethyl-2,3,4,5,6,6a,7,8,8a,10,11,12,13,14b-tetradecahydro-1H-picene-4a-carboxylate.

The molecular formula of Madecassoside is C48H78O20, and CAS Number is 34540-22-2. The molecular weight of Madecassoside is 975.1. The color of Madecassoside is white or off-white. Madecassoside is soluble in water and produces a colorless, clear solution.

However, Madecassoside is also soluble in Dimethylformamide (25 mg/ml), Dimethyl sulfoxide (10 mg/ml), Ethanol (5 mg/ml), and Phosphate-buffered saline (10 mg/ml) with pH 7.2.

Madecassoside sources

The primary source of madecassoside is Centella Asiatica which is commonly known as Indian pennywort. Centella Asiatica is an annual herbaceous plant. Madecassoside is the primary triterpene and was extracted from Centella Asiatica first time in 1948. Centella Asiatica is widely used for treating skin and vascular diseases in India and Southeast Asia. Centella Asiatica is a well-documented medicinal plant in different traditional therapeutic systems like Ayurveda, traditional African medicine, and traditional Chinese medicine. Leaves of C. Asiatica contain a high concentration of bioactive phytochemicals compared with petals and roots.

Constituents of Centella Asiatica Extract

The C. Asiatica leaf extract contains triterpenoid glycoside (saponin) compounds. Like asiaticoside and madecassoside and their analogous aglycones like Asiatic acid and madecassic acid

Apart from these four primary bioactive phytochemicals of the C. Asiatica plant, some other constituents are β-sitosterol, brahmoside, brahminoside, brahmic acid, betulic acid, centelloside, daucosterol, hexacosanol octanoate, isothankunoside, isobrahmic acid, kaempferol, oxyasiaticoside, quercetin, succinic acid, thankunoside, and vanillic acid.

A pharmacological research study evaluated the bioactive components in the titrated extract of Centella Asiatica and a total triterpenoid fraction of Centella Asiatica. These findings revealed that Asiatic acid (30%), madecassic acid (30%), asiaticosides (40%) were present in the titrated extract of Centella Asiatica(TECA). Total triterpenic fraction comprises asiatic acid and madecassic acid (60%) as well as asiaticosides (40%).

How does Madecassoside work?

Madecassoside has a wide range of skin benefits along with other multiple therapeutic properties like anti‑neuroinflammatory and antioxidative properties.

The anti-inflammatory activity of madecassoside obtains by inhibiting cyclooxygenase-2 (COX-2) and prostaglandin (PG) production.  Madecassoside significantly downregulates UV-induced melanin synthesis. Moreover, it also suppresses protease-activated receptor-2 expression and inhibits melanosome transfer to keratinocytes and melanocytes. Thus, Madecassoside is adequate to treat and prevent hyperpigmentation.

Madecassoside helps wound healing by following more than one mechanism of action. Like angiogenesis, antioxidative activity, and collagen synthesis.

Madecassoside Manufacturing Process

Madecassoside is extracted from the Centella Asiatica plant. The leaves, roots, and petioles are collected from glass house-grown Centella Asiatica. After collection of the plant parts, thoroughly wash with running tap water for complete removal of dirt. Then, all these plant parts are free-dried overnight at -200 C. Then, ground all the tissue parts using a grinder and add liquid nitrogen for sample homogenization before starting the Madecassoside extraction process.

Erlenmeyer flask and covered with Nescofilm required for extraction. The solvent is a mixture of methanol and water in a ratio of 9:1 v/v. The plant tissue sample and solvent ratio for extraction is 1:10 w/v. Then, the mixture of solvent and tissue needs to shake at 90 – 100 rpm for 30 h at room temperature. Whatman No. 1 paper is used for filtration. Then the filtrate is collected under reduced pressure. The collected filtrate is then stored in a screw-capped glass jar.

The filtrate is then evaporated to the dry, and the collected residue is re-dissolved in a mixture of methanol and water in a ratio of 9:1 v/v under reduced pressure. Then, microfuge tubes are used for sample collection and vortex for 2 – 3 min. Then, centrifuge the content to remove particulate matter, and supernatants are collected for HPLC. However, the sample is passed through bacteriological membranes (0.2 μm pore size) before HPLC. A Novapak C18 column is used for chromatographic separation.

Madecassoside Applications

Nowadays, madecassoside is added to a range of cosmetic preparations, including sunscreen, anti-aging, skin brightening, and sensitive skin care products. Madecassoside-containing sunscreen helps to reduce skin redness because of sunburn. This natural ingredient is also added to a face mask for an instant soothing effect. Individuals with rosacea or sensitive skin get to benefit from madecassoside-contained skincare products. Madecassoside with other ingredients improves skin tone and hydration. Therefore, users can get bright and calm skin. Madecassoside has potent antioxidant property that strengthens the skin and protects the skin barrier. Madecassoside has anti-inflammatory properties that are effective to reduce skin inflammation and aging symptom.  Madecassoside with other organic ingredients like a peach leaf in the formulation of clarifying body spray reduces blemishes, minimizes blackheads, regulates sebum production, and soothes skin.
