Madecassoside is a bioactive triterpene that is primarily obtained from Centella Asiatica plant extract. Asiaticoside A is another name for madecassoside. Chemically, madecassoside is also known as [6-[[3,4-dihydroxy-6-(hydroxymethyl)-5-(3,4,5-trihydroxy-6-methyloxan-2-yl)oxyoxan-2-yl]oxymethyl]-3,4,5-trihydroxyoxan-2-yl] 8,10,11-trihydroxy-9-(hydroxymethyl)-1,2,6a,6b,9,12a-hexamethyl-2,3,4,5,6,6a,7,8,8a,10,11,12,13,14b-tetradecahydro-1H-picene-4a-carboxylate.

The molecular formula of Madecassoside is C48H78O20, and CAS Number is 34540-22-2. The molecular weight of Madecassoside is 975.1. The color of Madecassoside is white or off-white. Madecassoside is soluble in water and produces a colorless, clear solution.

Madecassoside is extracted from the Centella Asiatica plant. The leaves, roots, and petioles are collected from glass house-grown Centella Asiatica. After collection of the plant parts, thoroughly wash with running tap water for complete removal of dirt. Then, all these plant parts are free-dried overnight at -200 C. Then, ground all the tissue parts using a grinder and add liquid nitrogen for sample homogenization before starting the Madecassoside extraction process. 

Erlenmeyer flask and covered with Nescofilm required for extraction. The solvent is a mixture of methanol and water in a ratio of 9:1 v/v. The plant tissue sample and solvent ratio for extraction is 1:10 w/v. Then, the mixture of solvent and tissue needs to shake at 90 – 100 rpm for 30 h at room temperature. Whatman No. 1 paper is used for filtration. Then the filtrate is collected under reduced pressure. The collected filtrate is then stored in a screw-capped glass jar. 

The filtrate is then evaporated to the dry, and the collected residue is re-dissolved in a mixture of methanol and water in a ratio of 9:1 v/v under reduced pressure. Then, microfuge tubes are used for sample collection and vortex for 2 – 3 min. Then, centrifuge the content to remove particulate matter, and supernatants are collected for HPLC. However, the sample is passed through bacteriological membranes (0.2 μm pore size) before HPLC. A Novapak C18 column is used for chromatographic separation.
